Single-cell multiomics sequencing reveals the functional regulatory landscape of early embryos

چکیده
Abstract Extensive epigenetic reprogramming occurs during preimplantation embryo development. However, it remains largely unclear how the drastic contributes to transcriptional regulatory network this period. Here, we develop a single-cell multiomics sequencing technology (scNOMeRe-seq) that enables profiling of genome-wide chromatin accessibility, DNA methylation and RNA expression in same individual cell. We apply method depict map mouse find remodeling facilitates reconstruction genetic lineages early embryos. Further, construct zygotic genome activation (ZGA)-associated reveal coordination among multiple layers, transcription factors repeat elements instruct proper ZGA. Cell fates associated cis-regulatory are activated stepwise post-ZGA stages. Trophectoderm (TE)-specific play dual roles promoting TE program while repressing inner cell mass (ICM) ICM/TE separation.

Background: Mammalian preimplantation development is a complex process involving dramatic changes in the transcriptional architecture. However, it is still unclear about the crucial transcriptional network and key hub genes that regulate the proceeding of preimplantation embryos. Materials and Methods: Through single-cell RNAsequencing (RNA-seq) of both human and mouse preimplantation embryos, ...
متن کاملSingle-Cell Multiomics: Multiple Measurements from Single Cells
Single-cell sequencing provides information that is not confounded by genotypic or phenotypic heterogeneity of bulk samples. Sequencing of one molecular type (RNA, methylated DNA or open chromatin) in a single cell, furthermore, provides insights into the cell's phenotype and links to its genotype. Nevertheless, only by taking measurements of these phenotypes and genotypes from the same single ...
متن کاملSingle-cell RNA sequencing reveals intrinsic and extrinsic regulatory heterogeneity in yeast responding to stress
From bacteria to humans, individual cells within isogenic populations can show significant variation in stress tolerance, but the nature of this heterogeneity is not clear. To investigate this, we used single-cell RNA sequencing to quantify transcript heterogeneity in single Saccharomyces cerevisiae cells treated with and without salt stress to explore population variation and identify cellular...
متن کاملthe effect of functional/notional approach on the proficiency level of efl learners and its evaluation through functional test
in fact, this study focused on the following questions: 1. is there any difference between the effect of functional/notional approach and the structural approaches to language teaching on the proficiency test of efl learners? 2. can a rather innovative language test referred to as "functional test" ge devised so so to measure the proficiency test of efl learners, and thus be as much reliable an...

Aged haematopoietic stem cells (HSCs) generate more myeloid cells and fewer lymphoid cells compared with young HSCs, contributing to decreased adaptive immunity in aged individuals. However, it is not known how intrinsic changes to HSCs and shifts in the balance between biased HSC subsets each contribute to the altered lineage output. Here, by analysing HSC transcriptomes and HSC function at th...
